3. 정제 조건

His-tagged gp96 protein was expressed in E. coli BL21 (DE3)and purified by nickel affinity chromatography and by gel filtration chromatography, and dialyzed against buffer (25mM Tris-HCl, pH7.5, 50 mM NaCl). 전에 한정민박사님이 위와 같은 방법으로 정제하여 실험하였다고 합니다. 논문을 참조하라고 하셔서 논문에 나온 정제방법을 알려드립니다.

4. 전해드릴 plasmid는 앞에 his -tag가 붙어있고 GP96 (22번에서 799번 AA까지 cloning 한 것 입니다.) pET28b 에 들어있습니다.
