Tranfection using Adenovirus method

1)    BAEC culture (50~60% confluent) in 6-well plate

2)    DNA 2mg + HBS 50ml + virus 20ml, 30min incubation, RT

3)    1mg/ml poly-L-lysine 100ml를 HBS 2.4ml에 가하여 섞고 2)에 50ml added.

이때 poly-L-lysine은 virus prep에 사용했던 것은 사용하지 말고 새로 만들어야 한다.

4)    30min incubation, RT

5)    0.5% FBS(HI) DMEM 1ml 가하여 virus-polylysine-DNA와 잘 섞어준다.

6)    BAECs을 PBS(or HBSS)로 wash후 PBS 2ml 가하여 1hr incubation

    37°C, 5.0% CO₂ incubator

7)    PBS를 제거하고 5)의 adenovirus-polylysine-DNA complex를 넣어준다.

8)    1hr incubation, 37°C, 5.0% CO₂ incubator

9)    10% FBS(HI) DMEM 1ml added in 6-well plate

10)  cell이 confluent해질 때까지 키운다. (24~36hrs)

필요하다면 media를 보충해 줄 수 있음.
