Peroxisome에 있는 PBE를 IP할 때 쓰는 lysis buffer. (Dr. Han)

 

· lysis buffer

        25mM Tris-HCl (pH 7.5)

        10mM NaCl                           +  Protease inhibiter

        미리 만들어 둘 것. 그리고,

        1% Triton X-100

 

                        ↓  Ice 30min

 

Sonication 15sec (cell일 경우)

    or

Homogenation ( tissue일 경우)

 

                         ↓ 

  

Centrifugation 18,000rpm ( 1hr )

 

         ↓                       ↓ 

       Sup               Pellet (버린다)

  

         ↓ 

 

Triton – Free buffer로 1/2 dilution (sup과 동일 vol을 섞어준다.)

 즉, 0.5% Triton X-100 이 된다.

 

 

 

l  Washing buffer

   25mM Tris-HCl (pH 7.5)

10mM NaCl

0.5% Triton X-100
